I pulled Dr. Darzynkiewicz's answer (below) to a post regarding sorting live proliferating cells from the archives (July 2007). Does anyone have a specific reference for this in action?? Looking thru the literature on this is daunting, to say the least.
"Cells that proliferate, especially in S and G2M, have higher RNA and
protein content and are distinctly larger than non-proliferating cells
The difference is particularly striking (~ 10 fold in RNA content) if
one compares quiescent genuine G0- with G1-S-G2M proliferating cells.
Cell size, thus (volume, forward light scatter) could provide some
discrimination between live cycling vs non-cycling cells, without a need
to for fluorochrome-labeling."
