Depending on your instrument performance and the size of the bugs you shoul=
d ideally sort from forward scatter versus green fluorescence as expression=
 is volume dependent. You will then be able to sort cells with the same exp=
ression intensity but different forward scatter as shown in the example fro=
m my tutorial or cells with different intensity but similar scatter. Howeve=
r if the investigator uses a plasmid, the expression intensity is less info=
rmative then using a single gene copy. Ideally you would want to measure ag=
ainst DNA content but the cells might not be too happy about that.
