[Cytometry] Indo-1 on an Aria?

Nick Goulding did flow calcium ion analysis in our lab a long time ago.  He developed a method for hanging a drop of the agonist on the side of the sample tube -- in such a way that it would only be released into the cell suspension when the tube was vortexed.

So -- put your cell suspension into a tube,
  hang the agonist solution on the side of the tube, 
 load the tube into the cytometer, 
start taking a minute or so of base line flow data,  
and then vortex the tube in place to start the calcium flux reaction.
