DNA free cytoplast sorting

With bacteria I can sort envelopes that can be distinguished by the
bis-oxonol uptake, reduced light scatter and the absence of DNA staining
by PI and EB. Any reason why you can't use PI?

After enucleation treatment, I wish to isolate DNA free ES cell
cytoplasts 
using FACS. I have two questions:

1. Can FACS be used to isolate cytoplasts? (by using FCS to distingish
the 
cytoplast from whole cells for example.)

2. How can I verify lack of DNA without exposing my cells to UV (cant
use 
PI)?
