T cell activation assay

Hi Flowers,

could anyone suggest the best method to assess T cell activation which does not require
long-term ex vivo culture? We try to characterize the impaired fuction of dendritic cells
(DCs) isolated from tumor-bearing mice. The problem is that cultured splenic DCs regain
their function during in vitro culture and the effects we are looking for become lost. We
tried also intracellular staining for IFNgamma on freshly isolated CD3+ cells, but
without additional costimulation the level of staining was very low. Is it posssible to
improve our ICS?
