Labeling Nuclear Antigens?

I would like to post a question for a colleague who is interested
in labeling a nuclear protein antigen that may also appear in the
cytoplasm.  Thus he needs to remove the cytoplasm and
permeabilize the nucleus.

He would like your advice on the best protocols to achieve this. 
Do the new "fix and perm" type reagents adequately permeabilize
the nuclear membrane?

Have there been major advances in this area since the older
protocols designed for total nuclear DNA measurements?

Any advice would be gratefully received, and if there is interest
we will post a summary of responses on this mailing list.
